Comparative Analysis of Mesenchymal Stem Cell Exosome Regenerative Potential in Wound Healing
This investigation examined the therapeutic promise of nano-sized vesicles secreted by mesenchymal stem cells (MSCs) sourced from bone marrow, adipose tissue, and umbilical cord. These exosomes carry vital bioactive molecules—including DNA fragments, microRNAs, proteins, and lipids—that orchestrate critical wound healing processes like cellular division, migration, and angiogenesis. Our findings demonstrate that all three MSC-derived exosome types contain essential regenerative mediators such as VEGF-A, FGF-2, HGF, and PDGF-BB. Notably, TGF-β emerged exclusively in umbilical cord MSC exosomes, highlighting origin-specific bioactivity. Experimental doses up to 20 μg showed dose-dependent enhancement of keratinocyte and fibroblast movement, with BMMSC exosomes driving strongest fibroblast activation and UCMSC exosomes optimizing keratinocyte responses. Clinical relevance suggests these exosome profiles could translate into novel biologic therapies for chronic wound management.
